Evaluation of Degradability of Aroclor 1242 by an Aceticlastic Methanogenic Consortium
ثبت نشده
چکیده
The rate and extent of degradation of Aroclor 1242 from contaminated surface soil was investigated, using aceticlastic mathanogenic consortia enriched from municipal anaerobic digester. Two different experiments, “intermediate feed” and “starve and feed” were conducted using triplicate 160-mL serum bottle microcosms, containing either 0.1 g, 1 g, 10 g, or 50 g of soil plus 100 mL of the inoculum. The methanogenic consortia in both the experiments were manipulated by feeding them with glacial acetic acid at different time intervals, during 30 days incubation. The fate of Aroclor 1242 in both the experiments was monitored using GC-ECD and GC-MSD analysis. Significant differences were observed between inoculated and uninoculated (control) microcosms, thus suggesting that methanogenic consortium might have been responsible for the degradation of Aroclor 1242. However, GC-MSD results could not confirm that disappearance of PCB was due to anaerobic dehalogenation. Another experiment was performed to evaluate if the removal of Aroclor 1242 was due to its evaporation. It was confirmed that disappearance of Aroclor 1242 was not due to evaporative losses during sample extraction. The actual reason for Aroclor 1242 disappearance from the surface soil microcosms could not be determined from this study.

Evidence for para dechlorination of polychlorobiphenyls by methanogenic bacteria.
When microorganisms eluted from upper Hudson River sediment were cultured without any substrate except polychlorobiphenyl (PCB)-free Hudson River sediment, methane formation was the terminal step of the anaerobic food chain. In sediments containing Aroclor 1242, addition of eubacterium-inhibiting antibiotics, which should have directly inhibited fermentative bacteria and thereby should have ind...
متن کاملDistinct phospholipases A2 regulate the release of arachidonic acid for eicosanoid production and superoxide anion generation in neutrophils.
Arachidonic acid (AA) released from membrane phospholipids by phospholipase A2 (PLA2) is important as a substrate for eicosanoid formation and as a second messenger for superoxide anion (O2-) generation in neutrophils. Different isoforms of PLA2 in neutrophils might mobilize AA for different functions. To test this possibility, we sought to characterize the PLA2s that are activated by the neutr...
متن کاملInfluence of aroclor 1242 concentration on polychlorinated biphenyl biotransformations in hudson river test tube microcosms.
When 93.3 to 933 (mu)mol of Aroclor 1242 per kg was added to Hudson River sediment test tube microcosms, the rates of polychlorinated biphenyl biotransformations increased with increasing Aroclor 1242 concentration after a 4- to 8-week acclimation period. In contrast, when 37.3 (mu)mol of Aroclor 1242 per kg was added, polychlorinated biphenyl biotransformations occurred at slow constant rates.
متن کاملNeutrophil function after exposure to polychlorinated biphenyls in vitro.
Polychlorinated biphenyls (PCBs) are known to be immunotoxic, yet the effects on neutrophil (PMN) function are not well characterized. We incubated PMNs isolated from rat peritoneum with a mixture of PCB congeners, Aroclor 1242, in the absence or presence of either phorbol myristate acetate (PMA) to stimulate generation of superoxide anion (O2-) or N-formyl-methionyl-leucyl-phenylalanine (fMLP)...
متن کاملEvaluation of cumulative PCB exposure estimated by a job exposure matrix versus PCB serum concentrations.
Although polychlorinated biphenyls (PCBs) have been banned in many countries for more than three decades, exposures to PCBs continue to be of concern due to their long half-lives and carcinogenic effects. In National Institute for Occupational Safety and Health studies, we are using semiquantitative plant-specific job exposure matrices (JEMs) to estimate historical PCB exposures for workers (n ...
